Optical super-resolution imaging is having a major impact in the area of biophysics and basic biology. The main reason for this development is the new access to scales on which macromolecular interactions are occurring within cells and related living systems. I will describe some of the optical and data processing considerations in designing a practical super-resolution imaging system based on the detection and localisation of single molecule fluorescence (often known as PALM/STORM). I will also describe applications that have allowed us to advance biophysical understanding of some cellular signalling mechanisms.
